How to tune up my bicycle

I'm going to have my bike assembled by a shop and I'm wondering if it's worth paying extra to have them "tune-up" the bike or just pay their standard bike assembly fee. Since it's new I would think it wouldn't need a tune up but this is my first time doing this so I'm not entirely sure. In most cases, all that isn't necessary. A shop will do that but charge you an arm and a prosthesis.
